E COLI for our BRAIN:

What'll they think of next? A protein from E Coli to alter Neural Progenitors Stem Cells by design into Specific Types of Neurons:
As the recipe book for turning stem cells into other types of cells keeps growing larger, the search for the perfect, therapeutically relevant blend of differentiation factors is revealing some interesting biology.
This new research reports the possibility that a protein from E. coli bacteria combined with small molecules can act synergistically to push pluripotent cells into functional neurons.
It seems that when it does the pushing will very much determine what sort of neuron we will that is pushed on stage
Pluripotent stem cells (PSCs) possess two important properties, including indefinite self-renewal and the ability to differentiate into most cell types
In all the tissues of our body, these pluripotent cells go down a well defined sequence of stages whereby they grow in number and various differentiations take place. Just when and how long each differentatied stage is allowed to last determines what sort of cell will emerge.
In the brain for example, at the simplest level all neurons and astrocytes emerge from a common pluripotent cell, and some of the astroctye progenitors seem to brancn off and form oligodendroctyes. And these each of those types develops still more different features from others of its class.
Accumulating evidence suggests that maintenance of pluripotency and differentiation into lineage-committed progenitors of PSCs are tightly regulated by the interplay of a few transcription factors termed master stemness regulators
As they go further down the road to those target destinations further key differentiations occur in sequence. and various of hese cells can become further kinds within that genus of cell. Until?
Until the cell type that they have become is called for by the brain and told to go "on stage" Before that they are held in a "pool" in a neurogenic niche "not yet ready for prime time"
These unique features of Pluripotent stem cells make them invaluable tools not only for exploitation as therapeutic agents in regenerative medicine but also for elucidation of the molecular mechanisms regulating proliferation and differentiation of stem cells
Much of what goes wrong in this long process from pluripotent cells to final "players" is due to the pushing of some of these cells either too soon or too late into their functioning roles.
In the case of skin tissue, we can get either insufficient wound healing or on the other hand, hypertrophic scarring, if the right time for coming on stage is not worked out by this complex system of which they speak
Here the possibility is raised that an Ecoli protein can be used to allow treating physicians to choose the desired time the release of a neurogenesis differentiation factor to either bring just the right actor on stage or perhaps prevent the wrong actor from going on.
The authors say that the differentiation of pluripotent stem cells can be conceived as two simple steps: first, a stem cell decides to no longer be a stem cell and begins to differentiate; second, the cell decides what kind of cell it wants to be.
"Although there has been considerable research in this field, there is still a bottleneck in being able to produce a high number of stem cells efficiently,"
Sox2 is a key player in the maintenance of pluripotency and stemness, and thus inhibition of its function would abrogate the stemness of pluripotent cells and induce differentiation into several types of cells
Here is striking new strategy that relies on a combination of Sox2 inhibition with lineage-specific induction to promote efficient and selective differentiation of pluripotent P19 cells into neurons.
The research began when Sungkyunkwan University scientists in Korea made a serendipitous discovery that Sox2—one of the four Yamanaka factors that affect a stem cell's ability to remain a stem cell or differentiate—can bind to a bacterial chaperone protein, Skp. A chaperone protein does very much that: it binds to a potentially active protein not allowing it to react until a later time.
They then tested what would happen if Skp was introduced into stem cells and found that it could initiate differentiation.
This led to the hypothesis that Skp could be combined with other techniques to make differentiation more efficient.
When P19 cells transduced with Skp protein, an inhibitor of Sox2, are incubated with a neurogenesis inducer, the cells are selectively converted into neurons that generate depolarization-induced sodium currents and action potentials
These authors developed a protocol to induce neuron differentiation, where the bacterial protein Skp acts in the first step by binding to Sox2 and inhibiting its function.
The small chemicals neurodazine (Nz) and neurodazole (Nzl) then act in the second step by telling the stem cell to become a neuron.
By influencing both steps, more functional neurons can be produced per batch of stem cells and at a faster rate if using either protein or small molecules alone. We are essentially deciding what happens within the 'womb" within each tissue where the pluripotent cells are produced, nurtured, differentiated one way or another, and then mobilized on stage to properly "born"
"The synergy thus mainly arises from combining suppression of stemness by protein and directing lineage-specific commitment by chemical inducers
Signaling pathway studies lead us to conclude that a combination of Skp with the neurogenesis inducer enhances neurogenesis in P19 cells by activating Wnt and Notch pathways. The present differentiation protocol could be valuable to selectively generate functionally active neurons from pluripotent cells.
One weakness, of course, of the protocol is that there are safety concerns around using bacterial proteins such as Skp in a therapeutic setting.
However, using this protein is advantageous compared to introducing genetic elements because protein cannot cause any genetic alteration or instability, which are the major concerns of using virus-mediated gene delivery to the stem cells.
The authors hope that this study can encourage others to develop similar approaches based on small molecule mimics of the first stage of stemness suppression.
Combining Suppression of Stemness with Lineage-Specific Induction Leads to Conversion of Pluripotent Cells into Functional Neurons
Shin says. "Hence this process stands as an example of rationally designed cell differentiation to achieve a high level of lineage commitment efficiency."
They are now working on using similar combinatorial approaches to explore how to make differentiation more efficient in other cell types, particularly those in the heart.

Super coiled DNA is far more dynamic than the 'Watson-Crick' double helix

There are actually about 3 billion base pairs that make up the complete set of DNA instructions in humans. This is about a metre of DNA. This enormous string of molecular information has to be precisely organised by coiling it up tightly so that it can be squeezed into the nucleus of cells.

To study the structure of DNA when it is crammed into cells, the researchers needed to replicate this coiling of DNA.

To investigate how the winding changed what the circles looked like, the researchers wound and then unwound the tiny DNA circles – 10 million times shorter in length than the DNA contained within our cells – a single turn at a time.

The researchers devised a test to make sure that the tiny twisted up DNA circles that they made in the laboratory acted in the same way as the full-length DNA strands within our cells, when it is referred to as ‘biologically active’.

They used an enzyme called ‘human topoisomerase II alpha’ that manipulates the twist of DNA. The test showed that the enzyme relieved the winding stress from all of the supercoiled circles, even the most coiled ones, which is its normal job in the human body. This result means that the DNA in the circles must look and act like the much longer DNA that the enzyme encounters in human cells.

Coiling the tiny DNA circles caused them to form a zoo of beautiful and unexpected shapes. “Some of the circles had sharp bends, some were figure-8s, and others looked like handcuffs or racquets or even sewing needles. Some looked like rods because they were so coiled,” said the author.

The cryo-electron tomography of the tiny DNA circles also revealed another surprise finding. Base pairs in DNA are like a genetic alphabet, in which the letters on one side of the DNA double helix only pair with a particular letter on the other side.

While the researchers expected to see the opening of base pairs – that is, the separation of the paired letters in the genetic alphabet – when the DNA was under-wound, they were surprised to see this opening for the over-wound DNA. This is because over-winding is supposed to make the DNA double helix stronger.

The researchers hypothesise that this disruption of base pairs may cause flexible hinges, allowing the DNA to bend sharply, perhaps helping to explain how a meter of DNA can be jammed into a single human cell.

Folk artist Kovan alias Sivadass, who was arrested on charges of sedition following his controversial songs demanding prohibition in the State, was granted conditional bail by the Principal Sessions Judge on Monday.

When the artist’s bail application came up for hearing, Principal Sessions Judge Aadhinathan granted it on the condition that he executed two sureties for Rs. 5,000 from two persons.

It may be recalled that the Madras High Court had earlier granted an interim stay on the two-day police custody of the People’s Art and Literary Association (PALA) member, allowed earlier by a lower court.

Though the Public Prosecutor sought five-day police custody, the Chief Metropolitan Magistrate (CMM) had granted two-day custody with a direction that the accused should be allowed to meet his advocate twice every day.

The artist was arrested in Tiruchi on October 30 by a special team from here. He was actively involved in street plays and CDs and consistently demanding the closure of the State-run liquor shops.

Earth’s Schumann Resonance Accelerating?

The Ancient Indian Rishis called 7.83 Hz the frequency of OM. It also happens to be Mother Earth’s natural heartbeat rhythm, known as the “Schumann Resonance.” According to Wikipedia, “Schumann resonances are global electromagnetic resonances, excited by lightning discharges in the cavity formed by the Earth’s surface and the ionosphere.”
For many years this resonance frequency has hovered at a steady 7.83 Hz with only slight variations. In June 2014 that apparently changed.Monitors at the Russian Space Observing System showed a sudden spike in activity to around 8.5 Hz. Since then, they have recorded days where the Schumann accelerated as fast as 16.5 Hz. (The graph is usually blue with some green, and no white.) At first they thought their equipment was malfunctioning, but later learned the data was accurate. Everyone was asking, what’s causing this intermittent spiking activity?
Is the Earth’s frequency speeding up? Since the Schumann frequency is said to be “in tune” with the human brain’s alpha and theta states, this acceleration may be why it often feels like time has sped up and events and changes in our life are happening more rapidly.

Error in GTW Gravitation:

"The locally measured energy of any particle or photon changes (is "red-shifted" or blue-shifted) as it climbs out or falls into a static gravitational field."
The measurement part is correct but this is a consequence of the time dilation at the point where the observation is made and has nothing to do with 'photons' which, as we see from QED, are a phenomena associated with the interaction of EM energy only.
The other error in the paper is the assumption that a photon can be red-shifted. This is not so. The emission can be measured as red-shifted but this is consequence of time dilation (either at the emitter or detector) and has nothing to do with the EM wave or photon dynamics. In QED, if the amplitude of the wave changes between the emitter and absorber then the number of photons also changes (the number of photons is proportional to the square of the amplitude of the wave).
See the two papers I posted recently for further clarification of these points or the thread entitled 'On the Granularity of Photons' for an earlier draft of my position (a later and more accurate draft, No.5, will be available next week).

The Most Overlooked Green Site Practice

As design engineers I believe that our tendency is to focus on post construction stormwater controls and best management practices. This is natural and to some degree warranted- after all we are usually hired to design a finished product and how it gets constructed is often left to the contractor. Couple that with the fact that the insurance companies and attorneys are always advising design professionals that "we are not responsible, in any way, for the means, methods, sequence, procedures, techniques, scheduling of construction" and it is understandable that the focus is on post construction stormwater management. However, because of that I think Erosion Prevention and Sediment Controls (EPSC) during construction are often overlooked as a green site design and planning technique. Give your erosion prevention and sediment controls a little love!

It might not be sexy but EPSCs or lack thereof can have a tremendous environmental impact. According to the EPA a typical, unmanaged construction site can lose approximately 35-45 tons or soil per acre in one year. By comparison, forest or farm land will lose 1 ton or less per acre over the same time period. Left unmanaged that soil can travel downstream and clog natural and man-made waterways, affect water supplies, damage aquatic life, and otherwise adversely impact adjacent property owners and waterways. In addition, bare soil can increase runoff velocities resulting in further erosion on site and downstream, increase runoff volumes causing flooding and reducing groundwater recharge and increase water temperatures negatively impacting aquatic species. Beyond that, if you ever want to upset the neighbor of one of your construction sites try dumping some dirty water or sediment on their property - it will definitely get their attention!

If you are applying LID (Low Impact Development) techniques to your site, it becomes even more critical to pay attention to the erosion and sediment controls. I have experienced this on some of the first LID projects that I designed and I saw otherwise good LID designs perform poorly because the contractor did not properly install and maintain erosion controls. With traditional stormwater controls much of the flow is directed to catch basins and pipes where it is easy to manage sediment and control flows. However, when the post construction design relies on vegetative practices such as swales and raingardens it is critical to prevent erosion rather than just control sediment. And it is equally important to make sure that infiltration practices don't become clogged and compacted during construction which can negatively impact their post construction performance. Many times these items are overlooked because LID techniques are new to the design and construction team.

So what can you do? Of course it is very site specific and you may have little or no control over construction phase activities, but there are some very basic tenants that you can apply that will do wonders for your erosion and sediment controls during construction.
  1. Disturb as little area as possible - the most basic thing that you can do to reduce soil erosion and sediment loss is to limit the amount of area that you disturb. You can do this by getting involved early in the planning process and working with the architect or planner to locate the building and infrastructure and being mindful of the natural slopes and general topography.
  2. Phase your grading operations - the common practice on most sites, especially smaller ones, is for the excavator to come in at the begging of the project, strip all of the topsoil in disturbed areas and grade the whole site to sub-grade. Much or all of the site is then left disturbed for the life of the construction project. By phasing the grading operations you can limit the amount of time the land is disturbed and therefore limit the amount of time that it can erode.
  3. Reduce slope gradients and lengths - steep and long slopes erode exponentially more than shorter flatter slopes. Try to keep slopes to 4:1 or less and less than 40' long. If slopes are steeper and/or longer they should be terraced, furrowed, serrated or stepped.
  4. Establish vegetation on disturbed areas - the best way to limit erosion and control sediment is to get vegetation established. A good stand of grass will beat silt fence any day. Here is a case where the color green really means something - a brown construction site will not perform as well as a green one!Unfortunately, the vegetation is often put off until the completion of the project rather than using it during construction to control erosion.
  5. Maintenance, maintenance, maintenance - don't wait for erosion controls to fail or for the local of state authorities to come calling before cleaning and maintaining erosion controls. Its always easier to maintain than it is to fix it once it gets out of hand.
The five techniques above are really just a sampling of what can be done to help prevent erosion and sediment loss, but I think that the key points are to be mindful of it in the design phase, stay on top of it and establish vegetation early. Traditionally the focus has been more on sediment control, yet it should be on erosion prevention. Doing that will limit environmental impact and improve the post construction performance of storm water controls.

CAPSULE ENDOSCOPY

Understanding Capsule Endoscopy

What is Capsule Endoscopy?

Capsule Endoscopy lets your doctor examine the lining of the middle part of your gastrointestinal tract, which includes the three portions of the small intestine (duodenum, jejunum, ileum). Your doctor will give you a pill sized video camera for you to swallow. This camera has its own light source and takes pictures of your small intestine as it passes through. These pictures are sent to a small recording device you have to wear on your body.
Your doctor will be able to view these pictures at a later time and might be able to provide you with useful information regarding your small intestine.
Why is Capsule Endoscopy Done?

Capsule endoscopy helps your doctor evaluate the small intestine. This part of the bowel cannot be reached by traditional upper endoscopy or by colonoscopy. The most common reason for doing capsule endoscopy is to search for a cause of bleeding from the small intestine. It may also be useful for detecting polyps, inflammatory bowel disease (Crohn’s disease), ulcers, and tumors of the small intestine.

As is the case with most new diagnostic procedures, not all insurance companies are currently reimbursing for this procedure. You may need to check with your own insurance company to ensure that this is a covered benefit.
How Should I Prepare for the Procedure?

An empty stomach allows for the best and safest examination, so you should have nothing to eat or drink, including water, for approximately twelve hours before the examination. Your doctor will tell you when to start fasting.

Tell your doctor in advance about any medications you take including iron, aspirin, bismuth subsalicylate products and other over-the-counter medications. You might need to adjust your usual dose prior to the examination.

Discuss any allergies to medications as well as medical conditions, such as swallowing disorders and heart or lung disease.

Tell your doctor of the presence of a pacemaker or defibrillator, previous abdominal surgery, or previous history of bowel obstructions in the bowel, inflammatory bowel disease, or adhesions.
Your doctor may ask you to do a bowel prep/cleansing prior to the examination.
What Can I Expect During Capsule Endoscopy?

Your doctor will prepare you for the examination by applying a sensor device to your abdomen with adhesive sleeves (similar to tape). The pill-sized capsule endoscope is swallowed and passes naturally through your digestive tract while transmitting video images to a data recorder worn on your belt for approximately eight hours. At the end of the procedure you will return to the office and the data recorder is removed so that images of your small bowel can be put on a computer screen for physician review.
Most patients consider the test comfortable. The capsule endoscope is about the size of a large pill. After ingesting the capsule and until it is excreted you should not be near an MRI device or schedule an MRI examination.
What Happens After Capsule Endoscopy?

You will be able to drink clear liquids after two hours and eat a light meal after four hours following the capsule ingestion, unless your doctor instructs you otherwise. You will have to avoid vigorous physical activity such as running or jumping during the study. Your doctor generally can tell you the test results within the week following the procedure; however, the results of some tests might take longer.
What are the Possible Complications of Capsule Endoscopy?

Although complications can occur, they are rare when doctors who are specially trained and experienced in this procedure perform the test. There is potential for the capsule to be stuck at a narrowed spot in the digestive tract resulting in bowel obstruction. This usually relates to a stricture (narrowing) of the digestive tract from inflammation, prior surgery, or tumor. It’s important to recognize obstruction early. Signs of obstruction include unusual bloating, abdominal pain, nausea or vomiting. You should call your doctor immediately for any such concerns. Also, if you develop a fever after the test, have trouble swallowing or experience chest pain, tell your doctor immediately. Be careful not to prematurely disconnect the system as this may result in loss of pictures being sent to your recording device.
Capsule endoscopy may also be called:
  • capsule enteroscopy
  • wireless capsule endoscopy
Capsule endoscopy allows for examination of the small intestine, which cannot be easily reached by traditional methods of endoscopy.

What type of diseases can be diagnosed with capsule endoscopy?

Capsule endoscopy continues to improve technically. It has revolutionized diagnosis by providing a sensitive (able to identify subtle abnormalities) and simple (non-invasive) means of examining the inside of the small intestine. Some common examples of small intestine diseases diagnosed by capsule endoscopy include:
  1. Angiodysplasias (collections of small blood vessels located just beneath the inner intestinal lining that can bleed intermittently and cause anemia)
  2. Small intestinal tumors such as lymphoma, carcinoid tumor, and small intestinal cancer
  3. Crohn's disease of the small intestine

What are the limitations of capsule endoscopy?

While the capsule provides the best means of viewing the inside of the small intestine, there are many inherent limitations and problems with its use, the most important of which is thatthe capsuledoes not allow for therapy. Other problems include:
  1. Abnormalities in some areas of the intestine are missed because of rapid transit of the capsule and blurred, uninterpretable photographs. The images can also be blurred by retained stool of food debris. Hence the importance of a good bowel preparation prior to the capsule endoscopy.
  2. At times, transit is so slow that the capsule examines only part of the small intestine before the battery fails. The battery life is about eight hours.
  3. If abnormalities are discovered that require surgical resection or further investigation, it may be difficult to determine where in the small intestine the abnormality is and thereby help direct therapy.
  4. If there are narrow areas due to scarring (strictures) or tumors in the small intestine, the capsule can get stuck in the narrow area and cause an obstruction of the intestine that requires surgical removal of the capsule. (For this reason, in patients who are suspected of having a stricture, a self-dissolving, dummy capsule is swallowed first. If the dummy capsule sticks, it can be seen on an x-ray of the abdomen and the location of the stricture determined. Because it dissolves with time, however, the obstruction will resolve without surgery, and the real capsule will not be swallowed.)
  5. Finally, reviewing the tens of thousands of photographs is very time consuming for the conscientious physician.

.
