Language, uniquely, makes us human

FOXP2 is our 'language gene' and has a unique sequence in humans.

We humans tend to consider ourselves apart from other species. But we’re not really so different. So what makes us unique? I’d say it’s language, though not everyone would agree.
Some people insist it’s our large brains, but dolphins have proportionally larger brains than we do. Some still contend our opposable thumbs set us apart, but koalas have two thumbs on each hand.
We walk on two legs? Yes, of course, but the feathered species do that too.
Some are insistent that our individuality as a species rests on the fact we can use tools, but many diverse vertebrate species are tool-users, including primates, elephants and birds. Even the veined octopus and certain ants and wasps have been observed using tools.
The answer, then, is… language. We uniquely have the ability to communicate complex and abstract ideas.
At first it was spoken language. Then, independently, several human cultures developed the written word – the means to communicate with others over thousands of miles or years.
Through language we have built civilisations, developed science and medicine, literature and philosophy. We do not have to learn everything from personal experience, because through language we can learn from the experience of others.
Language makes us human, and it’s encoded in our DNA.
The language gene
FOXP2, known as the “language gene”, has a unique sequence in humans. While other living mammals share identical amino acids at two key amino positions 303 and 325, these amino acids are different in humans (threonine to asparagine at amino acid 303 and asparagine to serine at amino acid 325).
Such substitution mutations occurred some time after we diverged from our common ancestor with the chimpanzee 4-8 million years ago.
We shared this unique FOXP2 protein sequence with both Neanderthals and Denisovans, from which we diverged somewhere in the region of 400,000 years ago.
Compared to these other hominids, humans have an additional mutation in a region that regulates FOXP2 gene expression. Was it this latest mutation in FOXP2 that ensured our survival through better communication, as other hominids went extinct?
This mutation was swiftly incorporated into the human genome at high frequencies during the last 50,000 years suggesting it carries a survival advantage. Studies to understand the effect of this most recent change in FOXP2 are currently underway.
The FOXP2 gene is involved in brain development, particularly those areas involved in vocal behaviour. FOXP2 is particularly important for animals, including songbirds such as finches, canaries and parrots that learn to sing by imitation.
In the songbird brain, FOXP2 expression is highest when birds are learning to sing. Reduction of FOXP2 expression in the brain of zebra finches at this critical period left birds unable to completely or accurately learn to sing.
In humans, FOXP2 mutations are associated with severe speech and language deficits known as developmental verbal dyspraxia – affecting both the ability to coordinate vocal muscles in speech and causing language comprehension difficulties. What a terrible, isolating condition that must be.
In language we find both truth and beauty; then, being human, we use it to argue about what is true and beautiful.
Language is fundamentally what makes us what we are. Would you disagree? If so, please, use your voice and let me know.

Sitting down for more than four hours a day increases men’s risk of developing chronic diseases such as diabetes and hypertension, a new study revealed.
Image: Yuri Arcurs/Shutterstock
Office workers, truck drivers and couch potatoes beware: a University of Western Sydney study has found that that men who spend more than four hours of each day sitting down are more likely to experience chronic disease such as cancer, diabetes, heart disease and high blood pressure.
Emma George, a PhD researcher from the UWS School of Science and Health, worked in collaboration with Professor Gregory Kolt, Dean of Science and Health at UWS, and Dr Richard Rosenkranz from the Department of Human Nutrition at Kansas State University to conduct the study of 63,048 Australian males aged 45-64.
Study participants from the NSW 45 and Up Study were asked to report on a range of health-related variables including the presence or absence of chronic diseases, and their daily sitting time. Sitting time was divided into four categories: less than four hours, four to six hours, six to eight hours, and more than eight hours.
Ms George says, compared with those who spent four hours or less sitting down each day, participants reporting higher amounts of sitting were much more likely to report serious health conditions.
"The rates of chronic diseases reported by the participants exponentially increased in proportion with the amount of time the participants spent sitting down,” says Ms George.
Independent of factors such as age, BMI and level of physical activity, the amount of time men spent sitting each day was significantly associated with chronic disease and diabetes – indicating that, despite how active people may be outside of work hours, the amount of time spent sitting during the day may still have a significant impact.
"Despite your levels of physical activity, the more time you spend sitting the less time your body has to stay active and expend energy,” says Ms George.
Ms George says the study is highly relevant to office workers and anyone whose daily job requires them to sit down for long periods of time, such as truck drivers.
“The results of this study suggest that there is potential for people to improve their overall health if they found more opportunities to move around during the day and reduce the amount of time spent sitting,” she says.
“People should consider ways that they can integrate movement into their daily routines. Perhaps arranging  ‘active’ meetings rather than a teleconference, or walk around during your lunch break rather than sitting at your desk.”
The research is part of the 45 and Up Study, the largest long-term study of ageing in Australia, involving more than 267,000 people. The results have been published in the International Journal of Behavioral Nutrition and Physical Activity.

'Junk DNA' regulates cancer gene

The University of New South Wales

Many cancers occur when genes that suppress tumours are not functioning properly, but the study found that what was once considered 'junk DNA' can re-activate these tumour suppressor genes.

Research revealing that genetic sequences once considered as "junk DNA" can regulate cancer genes could lead to the development of new cancer drugs to re-activate tumour suppressor genes.
Researchers led by UNSW's Associate Professor Kevin Morris have discovered a new mechanism,  which re-activate genes that have been switched off.
Many cancers occur when genes that suppress tumours are not functioning properly, allowing the cancer to grow unchecked. The study by the international team is published in the journal Nature Structural and Molecular Biology.
It reveals how a tumour suppressor gene called PTEN, which is shut down in many cancers including skin cancer, is controlled. And it shows how DNA sequences long considered “junk” can influence disease genes.
“We show that there are multiple layers of complexity in the regulation of this PTEN gene, but also that it might be possible to switch it back on in cancer,” Dr Morris says.
The researchers hope this would make tumours more sensitive to chemotherapy and prevent the development of resistance to cancer drugs.
Less than 2 per cent of DNA in the human genome is made up of genes, which carry the blueprint for the production of proteins. Most of the other 98 per cent of the genetic code - known as noncoding DNA – was dismissed in the past as “junk”, because it does not make proteins.
Some short stretches of noncoding DNA are called pseudogenes, because of the similarity of their sequences to known genes.
Dr Morris, of the School of Biotechnology and Biomolecular Sciences in the Faculty of Science, says their research shows that a pseudogene regulates the activity of the PTEN tumour suppressor gene, and the “big deal” is that they found the pseudogene uses two different mechanisms to do this.
“This is the first time bimodal functionality of a pseudogene has been shown,” he says.
He says the exciting prospect is that small molecules could be used to de-activate the pseudogene in cancer. “When you knock out the pseudogene you get activation of the tumour suppressor gene, which would be a good thing.”
Co-author, Per Johnsson, of the Karolinska Institute in Sweden, says: “This means that we might one day be able to reprogram cancer cells to proliferate less, become more normal, and that resistance to chemotherapy can hopefully be avoided.”
Much more research will be needed before this approach is available for clinical use.
Although the work was carried out on just one gene, it could be applicable to many more.
“There is speculation now that there is one pseudogene for each gene,” says Dr Morris.
Last year researchers in the ENCODE public consortium showed that most noncoding DNA is active: it is transcribed into RNA molecules, although mostly of unknown function.
The two mechanisms the pseudogene uses to regulate the PTEN gene are: the pseudogene’s RNA  controls whether PTEN is active and produces any RNA itself; it also controls whether the gene’s protein is made, or not, elsewhere in the cell.
The team includes researchers from the Karolinska Institute and the Scripps Research Institute in the US.

The problem with maths

The University of Sydney

Why aren’t Australian girls studying maths? Only 1.5% of girls go on to study science or maths for their HSC; 4.4% of boys undertake advanced maths with physics and/or chemistry components.
Image: Sergey Nivens/Shutterstock
The percentage of girls studying no maths for their HSC has more than doubled in the past decade, a report co-authored by University of Sydney researchers shows.
Honorary Professor John Mack from the Faculty of Education and Social Work and Barry Walsh examined data of all Year 8 students in NSW to show the proportion who go on to study maths-science subject combinations for their HSC.
The data revealed there was a substantial increase in the number of girls studying no maths at all in the HSC, and also showed a substantial drop overall in the number of boy and girl students undertaking at least one maths and one science subject in the HSC.
"The decline in maths and science participation coincided with the removal in 2001 of the HSC requirement for undertaking English and at least one course in maths or science," says Dr Rachel Wilson from the Faculty of Education and Social Work, who helped prepare the report.
"It is not a requirement in New South Wales, Victoria and Western Australia; although it is compulsory in SA, and to a small extent in Queensland and the Northern Territory. The study calls for policy change to make these subjects mandatory in order to lift participation in high school and to attract more girls to maths and science."
The figures show that in 2001, 9.5 percent of girls undertook no mathematics course for their HSC. In 2011, this figure was 21.8 percent - more than double that of 10 years previously.
Furthermore, in the same period the total proportion of Year 8 girls who went on to study intermediate or advanced maths dropped from 25 percent to 18 percent.
In terms of undertaking science and maths the study also showed that only 1.5 percent of girls and 4.4 percent of boys go on to study advanced maths with both physics and chemistry.
In 2001 some 19.7 percent of boys and 16.8 percent of girls studied a math-science combination in the HSC. In 2011 these figures had dropped to 18.6 percent of boys and 13.8 percent of girls. The decline has occurred despite the fact HSC participation increased by 5 percent over the period.
This analysis suggests there is an urgent need to address declining female participation and stagnated male participation in intermediate/advanced maths-and-science combinations of study.
The proportion of girls studying such combination subjects has dwindled since 2001 and there is now a greater gender disparity in maths/sciences participation than there was in the 1980s.

Konark Sun temple!!

Another lost technology of Konark Sun temple!!
The uniqueness of the temple lies in the fact that between every two stone pieces there lies an iron plate (this can be clearly seen). The temples higher floors have been reinforced using massive iron beams. This fantastic effort inhuman perseverance took 1200 workers about 12 years to complete and that the ’’Dadhinauti’’ (Peak) of the main temple had to be installed by the 12 year old son of the Chief Architect. The said peak being a 52 ton magnet. This magnet was the reason the entire edifice endured the harsh conditions (being on the sea front) for centuries without being affected. The main pratima (idol) was believed to be floating in the air because of the unique arrangements of the main magnets and other series of magnets. The placement of the temple had been aligned in a way that the first rays of the Sun falling on the coast would pass thru the Nata Mandir and would reflect from the diamond placed at the center of this idol in the Main Sanctum . This phenomena would last for a couple of minutesduring the early morning. These magnets were later removed by the Britishers for acquiring the magnetic stone. Brien Foerster Jason Martell

Another legends state that, the magnetic effects of the lodestone was so strong that it disturbed the ships compasses that passed by the coast and the ships would run aground. To save their trade and their ships, the Portuguese took away the lodestone. The lodestone that was acting as the central stone and keeping all the stones of the temple wall balanced, fell out of alignment because of its removal and eventually led to the destruction of Main Sanctum.

Jain Narayan Temple at Pattadakal, Karnatak (Rock cut Architecture)

Chalukyan rulers were not only empire builders, but great patrons of art whose encouragement prompted the artists and craftsmen to experiment and innovate in different architectural styles and giving it a new dimension. It is in their period that transition from rock-cut medium to structural temples took place.

Pattadakal located in Bijapur district of Karnataka was not only popular for Chalukyan architectural activities but also a holy place for royal coronation, 'Pattadakisuvolal'. Temples constructed here mark the blending of the Rekha, Nagara, Prasada and the Dravida Vimana styles of temple building.
The oldest temple at Pattadakal is Sangamesvara built by Vijayaditya Satyasraya (AD 697-733). The other notable temples at Pattadakal are the Kadasiddhesvara, Jambulingeswara both attributed to 7th century A.D. while Galaganatha temple was built a century later in the style of rekha nagara prasada. The Kasivisvesvara temple was the last to be built in early Chalukyan style. The Mallikarjuna temple was constructed by Rani Trilokyamahadevi to celebrate the victory over the Pallavas by Vikramaditya II. She is also credited to have built the Virupaksha temple influenced by the architecture of the Kailasanatha temple at Kanchipuram. The Virupaksha temple later served as a model for the Rashtrakuta ruler Krishna I (757 -783 A.D.) to carve out the great Kailasa at Ellora.
However, the last addition at Pattadakal was made during the reign of Rashtrakuta ruler Krishna II of the 9th century A.D. in form of a Jaina temple, locally famous as Jaina Narayana, with its two lower storeys functional.
The sculptural art of the early Chalukyas is characterised by grace and delicate details. The ceiling panels of the navagrahas, dikpalas, the dancing Nataraja, the wall niches containing Lingodbhava, Ardhanarisvara, Tripurari, Varahavishnu, Trivikrama bear ample testimony to the sculptor's skill as well as the cult worship in vogue. The narrative relief illustrating certain episodes from the Ramayana, Mahabharata, Bhagavata and Panchatantra fitted well with these grand religious edifices.
The Sangamesvara, Virupaksha and Mallikarjuna temples at Pattadakkal exhibit to a large degree the southerly elements in their vimanas, as crystallized in the contemporary Pallava temples.
The Sangamesvara, the earliest of the three, built by Chalukya Vijayaditya (697-733), is nearer the Pallava form in that it has no sukanasika, while the other two, which possess this, are the earliest of the Chalukyan type and its derivatives possessing this architectural member, as also does the Kailasa at Ellora. Both the Sangamesvara and the larger Virupaksha are similar to each other in being square on plan from the base to sikhara. The Virupaksha, built by the queen of Vikramaditya II (733-46), is the earliest dated temple with the sukanasika, being closely followed by the Mallikarjuna, built by another queen of the same king.
The main vimana of the Sangamesvara is of three storeys. The the lowermost storey is surrounded by two walls, the inner and outer, the second storey being an upward projection of the inner wall, while the outer wall encloses the covered circumambulatory round the sanctum.

The Virupaksha is a large complex consisting of a tall vimana with axial mandapas and peripheral sub-shrines round the court, enclosed by a wall with gopura-entrances in front and behind, all designed and completed at one time. As such, this is the earliest extant temple-complex in the Chalukyan series. The massive gopuras are also the earliest. The compound-wall of the complex, following the plan of the group itself, has on its coping kuta and said-heads, suggestive of a derivation from the Shore-temple at Mahabalipuram-a device which gives the impression of a lower storey when viewed from a distance.
The Mallikarjuna, built immediately after and close to the Virupaksha, is a smaller temple with a four-storeyed vimana with a circular griva and sikhara. It has more or less a similar plan.

Ancient Kailashnathar rock cut Temple carved out of single block of stone

Ancient Kailashnathar rock cut Temple carved out of single block of stone with "out of world" precision
Ellora Caves (Maharashtra) belong to the highest achievements of mankind both regarding the engineering skills and arts. Words can not describe the incredible richness of artistic and phylosophical messages emanated by thousands of sculptures, drawings and inscriptions in Ellora.
Most amazing creation here is Kailasanatha (Kailash Temple). This is large (84.1 m long and 29,3 m high - two times larger than Parthenon in Athens), free standing structure with extremely complex structural planning. It was carved over the years and in this time with extreme precision were removed 400,000 tonnes of granite rock. Temple was shaped to resemble the sacred Mount Kailash and originally was plastered and painted in bright colors.
This group of 34 monasteries and temples belongs to the last great cave temples.
Temples were built for three religions - 12 Buddhist temples and monasteries (630 - 700 AD), 17 Hindu shrines (550 - 780 AD) and 5 Jain temples (800 - 1000 AD).

Effect by generations of ancestors:

The influence or impact of ancestors of the past three generations is maximum i.e. up to the generation of our great grand parents.

The ancestors of up to the last seven generations have some sizeable influence.

The ancestors beyond the seventh generation do not affect us much. This is because the give-and-take account with the ancestors is fully completed or completed to a large extent in a given birth. Hence in the next birth, the give-and-take with ancestors of this present birth will be negligible if at all. If a sizeable give-and-take account remains, then we are born in the same family once again.

The above pertains to how far we are affected by our lineage (i.e. the vertical family tree). If we look at it horizontally (or laterally), we are affected by our ancestors up to our second cousins.

Ancestors can cause us problems in life due to various factors such as needing our help, to fulfill desires, settle give-and-take accounts, etc.

10 things you need to know about Stephen Hawking

1) Born in Oxford on January 8 1942 - 300 years after the death of astronomer Galileo Galilei - Professor Hawking grew up in St Albans, Hertfordshire. After being diagnosed with a rare form of motor neurone disease - amyotrophic lateral sclerosis (ALS) - at the age of 22, Hawking was given just a few years to live.

2) Hawking is as much a celebrity as he is a scientist, having appeared on The Simpsons, Star Trek and having provided narration for a British Telecom commercial that was later sampled on a Pink Floyd album.

3) He had a difficult time at the local public school and was persecuted as a "swot" who was more interested in jazz, classical music and debating than sport and pop. Although not top of the class, he was good at maths and "chaotically enthusiastic in chemistry". Hawking has said of his workload as an undergraduate at Oxford "amounted to an average of just an hour a day". He also said: "I'm not proud of this lack of work, I'm just describing my attitude at the time, which I shared with most of my fellow students. You were supposed to be brilliant without effort, or to accept your limitations and get a fourth class degree."Despite his workload confession, Hawking got a first and went to Cambridge to begin work on his PhD - but he was already beginning to experience the first symptoms of his illness, having fallen over twice for no reason during the last year of his undergraduate degree.

4) Hawking has credited his marriage in 1965 to Jane Wilde, a language student, as a turning point in his life at a time when he was unsure as to what the point of a degree was if he was to die soon. They went on to have three children - Robert, Lucy, and Timothy.

5) At a meeting of the Royal Society meeting, Hawking interrupted a lecture by renowned astrophysicist Sir Fred Hoyle to let him know that he had made a mistake. When asked how he knew there had been an error, Hawking replied: "Because I've worked them out in my head."

6) During the 1970s Hawking produced a stream of first class research, including probably his most important contribution to cosmology: the discovery of Hawking radiation, which allows a black hole to leak energy and gradually fade away to nothing.

7) In the 1980s, Professor Hawking and Professor Jim Hartle proposed a model of the universe which had no boundaries in space or time. The concept was described in A Brief History Of Time, which sold 25 million copies worldwide.

8) In February 1990 he left his wife of twenty five years to set up home with one of his nurses, Elaine Mason. The couple married in September 1995 but divorced in 2006.

9) Among some of his more unconventional theories, Professor Hawking has predicted the end of humanity - due to global warming, a new killer virus, or the impact of a large comet.

10) In 2009 he was awarded the Presidential Medal of Freedom, the highest civilian award in the United States. Hawking was the Lucasian Professor of Mathematics at the University of Cambridge for 30 years, taking up the post in 1979 and retiring on 1 October 2009. He is also a Fellow of Gonville and Caius College, Cambridge having held many other academic positions. In recent years, Professor Hawking has examined the relationship between science and religion, writing a 2010 book Grand Design, which argues that evoking God is not necessary to explain the origins of the universe.

Biochemist creates CO2-eating light that runs on algae | Watch how it works

Our atmosphere is filling up with CO2 and we seem to be the major cause of that. The generally accepted solution seems to be cutting back on emissions as quickly as possible, but implementing such cuts is problematic because everyone has to agree to do more, which essentially ends up costing a lot of time and money.
There is an alternative to such measures, though. Instead of relying entirely on cutting emissions, why don’t we start taking CO2 out of the atmosphere? That’s exactly what biochemist Pierre Calleja is trying to do, and his solution almost sounds too good to be true.
Calleja has developed a lighting system that requires no electricity for power. Instead it draws CO2 from the atmosphere and uses it to produce light as well as oxygen as a byproduct. The key ingredient to this eco-friendly light? Algae.
Certain types of algae can feed off of organic carbon as well as sunlight, and in the process produce carbohydrate energy for themselves as well as oxygen as a waste product.
Cajella’s lamps consist of algae-filled water along with a light and battery system. During the day the algae produce energy from sunlight that is then stored in the batteries. Then at night the energy is used to power the light. However, as the algae can also produce energy from carbon, sunlight isn’t required for the process to work. That means such lights can be placed where there is no natural light and the air will effectively be cleaned on a daily basis.
What isn’t discussed in the video is how much maintenance such a light needs. However, the good news is algae can also act as a biofuel once separated from the water, so even if the lights need a water change out every so often, the waste algae just forms another type of fuel where as the water can be recycled.

What is post-traumatic stress disorder?

A history of childhood trauma can make someone more at risk of developing PTSD.

People have probably always known about the psychological effects of experiencing life-threatening events such as military combat, natural disasters, or violent assault. Literature through the ages – some of it thousands of years old – provides many vivid portrayals of these internal struggles to recover from horrific experiences.
It was not until 1980, however, that these reactions were formally recognised by the international psychiatric community. The name chosen was post-traumatic stress disorder, or PTSD, and the diagnostic criteria were agreed.
Before discussing the nature and treatment of PTSD, it’s important to emphasise that human beings are generally resilient. Most people exposed to potentially traumatic events recover well with help from family and friends, and don’t develop mental health problems.
For those who don’t recover so well, PTSD is only one possibility, with depression, substance abuse, anxiety, and physical health problems also common. But PTSD is the only condition specifically tied to a traumatic experience.
SymptomsPTSD is a serious psychiatric disorder characterised by three groups of symptoms:
  1. Reliving the traumatic event. People with PTSD describe vivid, painful images and terrifying nightmares of their experience.
  2. Avoidance. People with PTSD try to avoid reminders of what happened. They become emotionally numb and socially isolated to protect themselves from the pain.
  3. Being constantly tense and jumpy, always on the look-out for signs of danger. PTSD is associated with significant impairment in social and occupational functioning.
Causes and risk factorsThe latest Australian National Mental Health Survey reported that over 4% of the population experienced the symptoms of PTSD in the last year.
The incidence of PTSD varies considerably depending on the type of trauma, with sexual assault consistently the highest (around half of rape victims will develop PTSD). Accidents and natural disasters – events that do not involve human malevolence – tend to be the lowest at around 10%.
About half the people who develop PTSD recover over the first six to twelve months. Unfortunately, in the absence of treatment, the other half are likely to experience chronic problems that may persist for decades.
So why do some people develop these problems and not others? The answer is a combination of what the person was like before the trauma, their experiences at the time, and what has happened since.
In terms of pre-trauma factors, genetic vulnerability plays a part, along with a history of trauma, particularly in childhood, as well as tendencies towards anxiety and depression. Not surprisingly, the more severe the traumatic experience (the higher the life threat or exposure to the suffering of others) the more likely the person will develop PTSD.
The final group of risk factors appear after the event, with the most important being social support: people who have a strong network of friends and family to support them after the experience are less likely to develop PTSD. Other life stressors during this period (such as financial, legal, health, or relationship problems) can also interfere with recovery.
Treatment
We have come a long way in improving treatments for PTSD and now have a large body of research evidence to guide our decisions.
The most effective treatment is trauma-focused psychological therapy. There are a few different forms, including cognitive behavioural therapies (CBT), as well as something called eye movement desensitisation and reprocessing (EMDR). The thing they share in common is providing the survivor with an opportunity to confront the painful memories, and to “work through” the experience in a safe and controlled environment. This therapy is not easy for either the patient or the therapist, but it is very effective in most cases.
Pharmacological treatment can also be useful in PTSD, especially in more complex cases and as an adjunct to trauma-focused psychological therapy. The most effective drugs for PTSD are the new generation anti-depressants – the selective serotonin re-uptake inhibitors or SSRIs. Other drugs can also be useful, depending on the clinical presentation.
The bottom line is that effective treatment is available if the PTSD sufferer can find their way to an experienced clinician.
We’ve come a long way in our understanding of mental health response to trauma in the last couple of decades, but many challenges lie ahead: Can we prevent the development of these problems? How should we respond with whole communities following widespread disaster such as bushfires, floods or terrorism? And can we improve the quality and availability of treatment?
As we address these challenges, we must strive to make sure the best possible care is available to those whose lives have been devastated by the experience of severe trauma.

Developing human kidney cells

Using human embryonic stem cells researchers produced human kidney cells. This advancement will have applications in toxicology, disease models and regenerative medicine.

Researchers at the Institute of Bioengineering and Nanotechnology (IBN) have successfully generated human kidney cells from human embryonic stem cells in vitro1. Specifically, they produced the renal cells under artificial conditions in the lab without using animals or organs. This has not been possible until now.
According to IBN Executive Director, Professor Jackie Y. Ying, “This discovery has wide-reaching implications for in vitro toxicology, drug screening, disease models and regenerative medicine. In particular, we are interested in applying our technology to develop predictive in vitro drug testing and renal toxicity models as alternatives to animal testing.”
IBN Team Leader and Principal Research Scientist Dr Daniele Zink elaborated, “The kidney is a major target organ for drug-induced toxic effects. Therefore, it is important for pharmaceutical companies to find out early in the development phase whether their drugs would cause nephrotoxicity in humans. However, animal models are of limited predictability, and there is currently no regulatory accepted in vitro assay based on renal cells to predict nephrotoxic effects. A major problem is the lack of suitable renal cells, which may now be resolved through our discovery.”
At present, human kidney cells are extracted directly from human kidney samples. However, this method is not efficient because such samples are limited, and the extracted cells die after a few cell divisions in the petri dish. Also, cells obtained from different samples would display variable features, depending on age, gender, health status and other conditions of the donor. Therefore, cells that have been isolated from human samples are of limited suitability for research and applications in industry and translational medicine, which require large cell numbers.
An alternative approach is to use human renal cell lines that have been rendered immortal, i.e. they can be reproduced indefinitely in the lab. However, such cells may not be used in many applications due to safety issues, and their functional features have usually been changed so profoundly that they may no longer be useful toward predicting cell behaviour in the human body.
IBN’s technique, on the other hand, enables human embryonic stem cells to differentiate into renal proximal tubular-like cells. This particular kidney cell type plays an important role in kidney disease-related processes and drug clearance. Results showed that the renal proximal tubular-like cells generated by IBN were similar to the renal proximal tubular cells isolated from fresh human kidney samples. For example, they displayed very similar gene and protein expression patterns. Also, since human embryonic stem cells may grow indefinitely in cell culture, the IBN researchers have discovered a potentially unlimited source of human kidney cells.
“We are currently adapting our approach to use induced pluripotent stem cells as the source,” shared Dr Karthikeyan Narayanan, IBN Senior Research Scientist. “We are also planning to modify our protocol in order to generate other renal cell types from stem cells.”
The IBN researchers have tested the renal cells they generated in in vitro nephrotoxicology models developed by the Institute, and have obtained very promising test results. They welcome industry partners to collaborate with IBN on commercializing this technology.
IBN has recently received a grant from A*STAR’s Joint Council Office Development Program to further develop predictive in vitro models for liver- and kidney-specific toxicity. This project will be conducted in collaboration with the Experimental Therapeutics Centre, the Bioinformatics Institute and the National University Health System.
